In 1826, Hampstead Academy was chartered by the state legislature. In 1827, the school was renamed Mississippi Academy, offering classes for both boys and girls. Subjects like rhetoric were taught in the school’s first building that contained four “handsome” rooms. A second building housed a chapel. In 1830, the school became Mississippi College, offering degrees in arts, sciences, and languages.

The school was given to the Presbyterian Church, although the church later suffered financial difficulty and returned the school to its original owners in 1850. It was then transferred to the Mississippi Baptist Convention making it the oldest Baptist institution in the country. During the Civil War, Mississippi College was able to keep its doors open by utilizing the campus for other purposes. Provine Chapel was used as a hospital and there are reports that the ground floor was used to stable horses owned by Ulysses S. Grant.
